Pi2, Pi9, Pi50, Pigm, Piz, Piz-t 同为Piz 位点上的复等位基因。其中,Fukunishiki 携带Pi2/Piz5,二八占携带Pi50,而Pi9来自小粒野生稻。

【定位与克隆】

Amante-Bordeos等首先将源自小粒野生稻中的抗稻瘟病基因导入栽培稻中,并命名为Pi9Pi9定位于水稻第6染色体,与之紧密连锁的RFLP标记有RG64和R2132。Pi2初步定位在水稻第6 染色体上RG64 和RZ612 之间,遗传距离分别是2.1 cM 和7.2 cM(Yu et al, 1991; Mew et al, 1994),后被精细定位在分子标记RG64 和AP22 之间,遗传距离分别为0.9 cM 和1.2 cM(吴金红等, 2002)。

【抗谱表现】

Pi9 抗谱相当广,对来自13 个国家的43 个稻瘟病菌株均表现出很高的抗性(Liu et al, 2002);Pi2 抗谱也相当广,对从中国收集的792 个稻瘟小种中的绝大部分表现抗性,只有7.55% 的小种能侵染携带Pi2 的亲本C101A51(Chen et al, 2001),此外,C101A51对来自13 个国家的43 个稻瘟病菌株中的36 个表现抗性(Liu et al, 2002)。

【基因的克隆与生物学功能分析】

Pi9 区域含有6 个串联排列的NBS-LRR 基因,其中Nbs2-Pi9Pi9Pi9 属于NBS-LRR 类基因,含有2 个内含子,长度分别是5362bp 和128bp,Pi-9 cDNA 全长4009 bp,包含3099bp 的编码区和910-bp 3' UTR。Pi-9 编码一个由1032 氨基酸组成的蛋白产物,该产物羧基端含有富亮氨酸重复域(LRRs),由17 个不完整的LRR 重复组成;在氨基端含有NBS 域,NBS 域内有激酶1a、2和激酶3a等结构(Qu et al., 2006)。

Pi9 在抗病植株中组成性表达,表达不受稻瘟病侵染诱导。Pi9 至少对21 个稻瘟病菌小种具有抗性,是一个稻瘟病广谱抗性基因。Pi9 对稻瘟病的广谱抗病性的分子机制可能是Pi9 可以识别不同稻瘟病菌小种的保守分子,或者Pi9 可以识别不同稻瘟病菌小种的不同分子。

Pi2 属于NBS-LRR类基因,含有2个内含子,长度分别是3839bp 和116bp,Pi2 cDNA 全长3332 bp,包含117-bp 5' UTR 和116-bp 3' UTR。Pi2 编码产物包含1032 个氨基酸,含有1 个核苷酸结合位点(NBS)和3 个富亮氨酸重复区(LRR);Pi2Pi-zt 的编码产物仅在3个LRRs区域有8个氨基酸的差异;而在这8突变氨基酸中,仅有1个位于xxLxLxx基序中(Zhou et al., 2006)。
